The first settlers came to Lewisberry in the 1730s, and these became some of the first Quaker settlements in America west of the Susquehanna River. The Quakers lived peacefully among the large Susquehannock Native American population.
During the American Revolution, Lewisberry became an important gun manufacturing site. Eli Lewis as the founder of the borough, and after his wealth grew because of the gun factory, he owned a considerable portion of the land in the area.
